I had to do an emergency install of 10.1 on my laptop (Toshiba Satellite 1905-303S) the road (system was Ubuntu; SuSE 10.1 was the most familiar distro I could finde in Frye's!). I merely set Jpilot for /dev/pilot and make sure I press the hot-sync button on the cradle (Tungsten E2) and wait a couple of seconds before I click on the hot-sync icon on Jpilot (0.99.9). Works every time. /dev/ttyUSB0 and 1 don't appear in the /dev directory until I press the hot-sync putton on the cradle. Try that approach, if you haven't already...
